Description

This Human Complement 3 (C3) ELISA Kit was designed for the quantitative measurement of Human Complement 3 (C3) protein in serum, plasma, cell culture supernates, urine, saliva. It is a Sandwich ELISA kit, its detection range is 4.7 ng/mL-300 ng/mL and the sensitivity is 4.031 ng/mL.

Linearity

To assess the linearity of the assay, samples were spiked with high concentrations of human C3 in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

Typical Data

These standard curves are provided for demonstration only. A standard curve should be generated for each set of samples assayed.

Materials provided
  • A micro ELISA plate --- The 96-well plate has been pre-coated with an anti-human C3 antibody.
  • Two vials lyophilized standard --- Dilute a bottle of the standard at dilution series, read the OD values, and then draw a standard curve.
  • One vial HRP-conjugated C3 antibody(100 x concentrate)(120 μl/bottle) --- Bind to the C3 in the samples or standards and react with the substrate to make the solution chromogenic.
  • One vial Wash Buffer (25x concentrate) (20 ml/bottle) --- Wash away unbound or free substances.
  • One TMBSubstrate (10 ml/bottle) ---Interact with HRP to elicit a chromogenic reaction.
  • One vial Stop Solution (10ml/bottle) --- Stop the color reaction. The solution color immediately turns from blue to yellow.
  • Four Adhesive Strips (For 96 wells) --- Cover the microplate when incubation.
  • An instruction manual

C3 plays a central role in the activation of the complement system. Its processing by C3 convertase is the central reaction in both classical and alternative complement pathways. After activation C3b can bind covalently, via its reactive thioester, to cell surface carbohydrates or immune aggregates.; Derived from proteolytic degradation of complement C3, C3a anaphylatoxin is a mediator of local inflammatory process. In chronic inflammation, acts as a chemoattractant for neutrophils. It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes.; Acts as a chemoattractant for neutrophils in chronic inflammation.; adipogenic hormone that stimulates triglyceride (TG) synthesis and glucose transport in adipocytes, regulating fat storage and playing a role in postprandial TG clearance. Appears to stimulate TG synthesis via activation of the PLC, MAPK and AKT signaling pathways. Ligand for C5AR2. Promotes the phosphorylation, ARRB2-mediated internalization and recycling of C5AR2.
