IL-1 receptor antagonist is a key endogenous regulator of IL-1 signaling in equine inflammatory biology, and this recombinant horse IL1RN provides a tag-free, full-length mature protein (26–177 aa) that faithfully reproduces the native sequence critical for competitive binding at the IL-1 receptor. Functional validation confirms an ED50 of less than 3.0 μg/ml in an IL-1α–dependent D10S proliferation inhibition assay (specific activity >333 IU/mg), demonstrating signaling potency appropriate for cell-based functional studies such as IL-1–driven proliferation blockade, NF-κB pathway dissection, and dose–response standardization as a reference in equine cytokine detection assays. Endotoxin levels below 1.0 EU/μg minimize LPS-driven artifacts, which supports use in sensitive immune cell activation and differentiation experiments as well as in vivo equine inflammation models. Purity exceeding 95% by SDS-PAGE, combined with this low endotoxin profile, also provides a suitable basis for antibody development workflows including immunization, ELISA coating, and western blot controls.
