  • Overlay Peak curve showing Hela cells stained with CSB-MA617930A0m (red line) at 1:100. The cells were incubated in 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 1h at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/100 dilution for 30min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

Mice were immunized with the recombinant human MBTPS1 protein (187-400aa), and B cells were isolated from their spleens. These B cells were then fused with myeloma cells to generate hybridomas. After screening and selecting, the MBTPS1 antibody-secreting hybridomas were identified and then cultured to obtain the MBTPS1 monoclonal antibody. The MBTPS1 monoclonal antibody is highly specific for human MBTPS1 protein and can be used in three applications including ELISA, WB, and FC. This antibody is purified using protein G affinity chromatography and has a purity of over 95%.

MBTPS1, also known as Site-1 protease (S1P), is important for maintaining lipid homeostasis in cells. It plays a crucial role in regulating the processing of a variety of transmembrane proteins in the endoplasmic reticulum (ER), including SREBP (sterol regulatory element-binding protein) transcription factors, which are key regulators of cholesterol and fatty acid metabolism.

Serine protease that cleaves after hydrophobic or small residues, provided that Arg or Lys is in position P4: known substrates are SREBF1/SREBP1, SREBF2/SREBP2, BDNF, GNPTAB, ATF6 and ATF6B. Cleaves substrates after Arg-Ser-Val-Leu (SREBP2), Arg-His-Leu-Leu (ATF6), Arg-Gly-Leu-Thr (BDNF) and its own propeptide after Arg-Arg-Leu-Leu. Catalyzes the first step in the proteolytic activation of the sterol regulatory element-binding proteins (SREBPs) SREBF1/SREBP1 and SREBF2/SREBP2.
