IL-12, the heterodimeric cytokine formed by IL-12B (p40) and IL-12A (p35), plays a central role in bridging innate and adaptive immunity by driving Th1 polarization, NK cell activation, and IFN-γ production. This recombinant human IL-12 heterodimer, expressed in mammalian cells and spanning residues 23–328 (IL-12B) and 23–219 (IL-12A) with C-terminal 10×His and Flag tags, preserves the native subunit pairing required for receptor engagement and downstream JAK2/TYK2–STAT4 signaling studies. Functional ELISA confirms binding to an anti-IL-12/IL-23 recombinant antibody with an EC₅₀ of 1.042–1.545 ng/mL, supporting use as a coating antigen for antibody development and validation or as a reference standard in cytokine detection assays. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the stringent criteria needed for PBMC and primary T-cell activation assays, where LPS-driven artifacts could otherwise confound cytokine-attributable responses.
