The DNA fragment encoding the human IL12 complex composed of partial human IL12B (Ile23-Ser328) with a C-terminal 10xHis-tag and the mature form of human IL12A ( Arg23-Ser219) with a C-terminal Flag-tag was co-expressed in the mammalian cells. The resultant product is the recombinant human IL12A&IL27B heterodimer protein. It is low in endotoxin, less than 1.0 EU/µg protein determined by the LAL method. The purity of this protein is greater than 95%. This recombinant IL12A&IL27B heterodimer protein has been validated to be biologically active through its interaction with anti-IL12/IL23 recombinant antibody (CSB-RA011587MA1HU). The EC50 for this effect is 1.042-1.545 ng/mL.
IL12, a heterodimer composed of IL12A and IL12B subunits, is a pleiotropic cytokine that targets T cells, NK cells, and bone marrow progenitor cells. IL12 can promote the proliferation of activated T cells and NK cells, enhance the cytotoxicity of T cells and NK cells, and induce the production of IFN-γ and TNF-β. IL12 plays an important role in the anti-tumor and in the balance of Th1/Th2 responses. The critical role of IL12 in anti-tumor immunity is linked to the innate resistance regulated by macrophages and NK cells and the adaptive immunity mediated by Th1 cells and CTL cells. As the research of IL12 antibody drugs becomes hot, the preparation of IL12 protein will promote the development of IL12-related drugs.
