Buffer

Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Description

NTMT1 catalyzes the trimethylation of N-terminal α-amines on X-Pro-Lys motifs, a modification implicated in DNA damage repair and mitotic chromosome segregation. This recombinant human NTMT1 covers the full-length mature protein (aa 2–223) and carries an N-terminal GST tag, positioning the fusion partner away from the catalytic domain to preserve active-site accessibility for substrate engagement. Functional validation confirms binding activity against immobilized CCL5 (2 μg/ml) with an EC50 of 3.029–5.404 μg/ml, supporting its use as a positive control in enzyme-linked binding assays and providing a suitable basis for inhibitor screening and IC50 determination. Purity exceeding 85% by SDS-PAGE meets the quality thresholds commonly required for enzymatic activity assays, substrate specificity profiling, and kinetic parameter analysis including Km and kcat measurements.

Function

Distributive alpha-N-methyltransferase that methylates the N-terminus of target proteins containing the N-terminal motif [Ala/Gly/Pro/Ser]-Pro-Lys when the initiator Met is cleaved. Specifically catalyzes mono-, di- or tri-methylation of the exposed alpha-amino group of the Ala, Gly or Ser residue in the [Ala/Gly/Ser]-Pro-Lys motif and mono- or di-methylation of Pro in the Pro-Pro-Lys motif. Some of the substrates may be primed by NTMT2-mediated monomethylation. Catalyzes the trimethylation of the N-terminal Gly in CENPA (after removal of Met-1). Responsible for the N-terminal methylation of KLHL31, MYL2, MYL3, RB1, RCC1, RPL23A and SET. Required during mitosis for normal bipolar spindle formation and chromosome segregation via its action on RCC1.
