Sonic hedgehog signaling governs critical patterning events in embryonic development and drives proliferative pathways exploited in multiple cancer contexts. This tag-free recombinant mouse Shh fragment (aa 26–198) encompasses the biologically active N-terminal signaling domain and demonstrates confirmed potency with an ED50 of 0.5–1.0 μg/ml in alkaline phosphatase induction assays using C3H10T1/2 cells, supporting use in osteogenic differentiation studies, stem cell fate commitment experiments, and receptor-ligand binding or competition assays. The E. coli expression system yields a non-glycosylated protein that provides a suitable basis for mechanistic studies where glycan-independent Hedgehog–Patched interactions are being interrogated, as well as for in vivo tumor biology models requiring defined ligand inputs. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based proliferation assays and sensitive downstream functional readouts.
