  • Immunofluorescence staining of PC-3 cells with CSB-PA00135A0Rb at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).

Sample type: Mouse Mouse kidney

Review: Immunostaining for H + -ATPase G1 subunit (ATP6V1G1) in mouse kidney showed expression in the cortical and medullary portion of the kidney. Staining was found in both the proximal tubules and distal nephron segments, with pronounced intercalated cell (IC) staining in both cortex and medulla. Gray stippled lines demarcate the zone of the inner medulla.

Function
Subunit of the V1 complex of vacuolar(H+)-ATPase (V-ATPase), a multisubunit enzyme composed of a peripheral complex (V1) that hydrolyzes ATP and a membrane integral complex (V0) that translocates protons. V-ATPase is responsible for acidifying and maintaining the pH of intracellular compartments and in some cell types, is targeted to the plasma membrane, where it is responsible for acidifying the extracellular environment. In aerobic conditions, involved in intracellular iron homeostasis, thus triggering the activity of Fe(2+) prolyl hydroxylase (PHD) enzymes, and leading to HIF1A hydroxylation and subsequent proteasomal degradation.
