Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4℃ for up to one week.

Target Background

Function
The master regulator for adhesive curli fimbriae expression; necessary for transcription of the csgBAC/ymdA operon. Plays a positive role in biofilm formation. May have the capability to respond to starvation and/or high cell density by activating csgBA transcription. Low-level constitutive expression confers an adherent curli fimbriae-expressing phenotype, up-regulates 10 genes and down-regulates 14 others.
Gene References into Functions
  1. Regulation of the master biofilm regulator, CsgD, was diverse, and the screen revealed several proteins and small RNAs (sRNA) that regulate csgD messenger RNA (mRNA) levels. Using previously published studies, we found minimal overlap between the genes affecting curli biogenesis and genes known to impact swimming or swarming motility, underlying the distinction between motile and sessile lifestyles. PMID: 29088115
  2. Promoter mutations in csgD that alter sigma factor utilization from RpoS to RpoD provide a unique mechanism for increasing biofilm formation, but the transition also disrupts normal rpoS-dependent CsgD regulation. PMID: 28406080
  3. RprA can directly reduce csgD mRNA levels and translation, and ce transcription of csgD. PMID: 22356413
  4. The authors propose that the cell motility for planktonic growth is repressed by CsgD, thereby promoting the switch to biofilm formation. PMID: 21421764
  5. Five different transcription factors interact to regulate the CsgD promoter. PMID: 20466769
  6. CsgD activation of glyA requires the participation of both known regulatory proteins, MetR and PurR PMID: 15644921
  7. role of the interplay OmpR and CpxR in affecting the transcription of the curli gene in response to variation of the medium osmolarity; CpxR represses csgD expression in high-salt-content medium, resulting in low curli production PMID: 15743952
  8. Our results indicate that CsgD can modulate cellulose biosynthesis through activation of the yoaD gene. In addition, the positive effect of CsgD on biofilm formation might be enhanced by repression of the fecR gene. PMID: 16513732
