Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

Expressing the recombinant Escherichia coli O6:K15:H31 (strain 536 / UPEC) ISCS protein generally involves constructing a plasmid encoding the Escherichia coli O6:K15:H31 (strain 536 / UPEC) ISCS protein (1-404aa). This plasmid is introduced into e.coli cells, followed by the selection and culturing of positive e.coli cells, induction of protein expression, and subsequent cell lysis. The protein is fused with a N-terminal 10xHis-SUMO tag and C-terminal Myc tag. The recombinant Escherichia coli O6:K15:H31 (strain 536 / UPEC) ISCS protein is purified through affinity purification, and SDS-PAGE analysis is conducted to confirm the presence of the protein and determine its purity. Function

Master enzyme that delivers sulfur to a number of partners involved in Fe-S cluster assembly, tRNA modification or cofactor biosynthesis. Catalyzes the removal of elemental sulfur and selenium atoms from cysteine and selenocysteine to produce alanine. Functions as a sulfur delivery protein for Fe-S cluster synthesis onto IscU, an Fe-S scaffold assembly protein, as well as other S acceptor proteins. Also functions as a selenium delivery protein in the pathway for the biosynthesis of selenophosphate.
