Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

Amino acids 1-423 constitute the expression domain of recombinant Human FBXL2. This FBXL2 protein is expected to have a theoretical molecular weight of 63.1 kDa. This FBXL2 recombinant protein is manufactured in e.coli. The N-terminal 6xHis-SUMO tag was fused into the coding gene segment of FBXL2, making it easier to detect and purify the FBXL2 recombinant protein in the later stages of expression and purification.

The human F-box/LRR-repeat protein 2 (FBXL2) is a member of the F-box protein family, serving as a substrate recognition component of the SCF (Skp1-Cul1-F-box protein) ubiquitin ligase complex. FBXL2 contains leucine-rich repeat (LRR) motifs, facilitating its interaction with specific substrates for ubiquitination and subsequent degradation by the proteasome. FBXL2 is involved in the regulation of diverse cellular processes, including cell cycle progression, signal transduction, and apoptosis. Its substrates include key proteins such as β-catenin and SREBP (sterol regulatory element-binding protein). Research on FBXL2 aims to elucidate its roles in cellular homeostasis, and development, and its potential implications in various diseases, including cancer and metabolic disorders.

Target Background

Function
Calcium-activated substrate recognition component of the SCF (SKP1-cullin-F-box protein) E3 ubiquitin-protein ligase complex, SCF(FBXL2), which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. Unlike many F-box proteins, FBXL2 does not seem to target phosphodegron within its substrates but rather calmodulin-binding motifs and is thereby antagonized by calmodulin. This is the case for the cyclins CCND2 and CCND3 which polyubiquitination and subsequent degradation are inhibited by calmodulin. Through CCND2 and CCND3 degradation induces cell-cycle arrest in G(0). SCF(FBXL2) also mediates PIK3R2 ubiquitination and proteasomal degradation thereby regulating phosphatidylinositol 3-kinase signaling and autophagy. PCYT1A monoubiquitination by SCF(FBXL2) and subsequent degradation regulates synthesis of phosphatidylcholine, which is utilized for formation of membranes and of pulmonary surfactant.
