Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

 Q&A
Q:

We previously purchased CSB-BP883633HU (lot# 03748), wanted to order again, but we were not overly happy with the concentration and purity of our previous order.
If we choose to order again, would you be able to guarantee a higher concentration, and higher purity? We are hoping for a concentration of ~2mg/mL, and purity of >95%.
Would these be possible?
Thanks so much!

A:
Thanks for your inquiry.
For your special request (Re-order for lot# 03748; Concentration ~2mg/mL; Expected purity >95%), our reply is listed as below.
This concentration is a bit difficult for Baculovirus system. The concentration of proteins expressed by Baculovirus are generally shipped in the range of 0.1-0.5mg/ml.
We can try to optimize the concentration to reach your expected concentration of 2mg/ml, but it's not 100% guaranteed.
As for purity, we can optimize the purity as much as possible, such as AKTA-SEC secondary purification, etc., but whether to carry out AKTA-SEC secondary purification, depending on the specific situation.
If the initial purification of the protein has reached the quality control standard (Purity≥85%), we will not carry out AKTA-SEC purification;
If you still require AKTA-SEC secondary purification to further improve the purity, then this requires an additional fees, and the lead time will be extended by 3-5 working days. (we only try to increase the purity on the basis of 85%, we can't guarantee 95% purity.)
Any more questions, please feel free to contact us.

Target Background

Function
This form constitutes the precursor of the pore-forming protein: upon cleavage, the released N-terminal moiety (Gasdermin-C, N-terminal) binds to membranes and forms pores, triggering cell death.; Pore-forming protein that causes membrane permeabilization and pyroptosis. Released upon cleavage and binds to membrane inner leaflet lipids. Homooligomerizes within the membrane and forms pores of 10-15 nanometers (nm) of inner diameter, triggering pyroptosis. The functional mechanisms and physiological proteases that cleave and activate this pore-forming protein are unknown (Probable).
Gene References into Functions
  1. Expression of GSDMC is increased by UV irradiation in human skin keratinocytes. GSDMC induction contributes to the increase of MMP-1 expression specifically via activation of ERK and JNK pathways. PMID: 29428815
  2. Gasdermin C is upregulated by inactivation of Tgfbr2 in the presence of mutated Apc, promoting colorectal cancer cell proliferation. PMID: 27835699
  3. Study investigated the expression pattern of the GSDM family genes in the upper gastrointestinal epithelium and cancers. PMID: 19051310
Subcellular Location
[Gasdermin-C]: Cytoplasm, cytosol.; [Gasdermin-C, N-terminal]: Cell membrane; Multi-pass membrane protein.
Protein Families
Gasdermin family
Tissue Specificity
Expressed mainly in trachea and spleen. In the esophagus, expressed in differentiating cells and probably in differentiated cells. Also detected in gastric epithelium.
