MICB functions as a stress-induced ligand for the activating receptor NKG2D on natural killer cells and CD8+ T cells, making it a critical node in immune surveillance and tumor immunology. This mammalian-expressed protein spanning residues 23–309 captures the complete extracellular domain architecture required for physiologically relevant NKG2D engagement, and its binding to an anti-MICA/B recombinant antibody yields an EC50 of 0.9180–1.062 ng/mL in functional ELISA, demonstrating the conformational integrity necessary for receptor-ligand interaction studies and therapeutic antibody epitope mapping. The quantified binding kinetics support its use in competitive inhibition assays to screen blocking antibodies, in SPR or BLI experiments for affinity characterization of NKG2D-targeting biologics, and as a positive control in immune checkpoint research focused on NK cell activation pathways.
