Target Background

Function
Cleaves aggrecan, a cartilage proteoglycan, at the '1691-Glu-|-Leu-1692' site (within the chondroitin sulfate attachment domain), and may be involved in its turnover. Has angiogenic inhibitor activity. Active metalloprotease, which may be associated with various inflammatory processes as well as development of cancer cachexia. May play a critical role in follicular rupture.
Gene References into Functions
  1. The developmental arrest of early secondary follicles following furin disruption might be rooted in the loss of the mature form of ADAMTS1 and compromised proliferation of granulosa cells in mutant mice. PMID: 28569793
  2. Data show that NOTCH1 is a target of ADAMTS1 metalloproteinase activity, which reduces Notch signaling, leading to increased satellite cell activation. PMID: 28939843
  3. Reduced tumors in ADAMTS1 knockout mice displayed a paradoxical increase in vascular density and vascular-related genes; a detailed characterization revealed an impaired vasculature, along with a minor infiltration of macrophages. melanoma xenografts showed a relevant induction of its expression in stroma compartments. PMID: 27120788
  4. Adamts1 acts as an extracellular matrix 'modifier', with miR-181d-induced downregulation, that regulates adipocyte lineage commitment and obesity. PMID: 27447109
  5. Genetic haploinsufficiency of Adamts1 in mice causes thoracic aortic aneurysms and dissections similar to Marfan syndrome. PMID: 28067899
  6. An ADAMTS1 blocking antibody suppressed the SPARC-induced collagen I secretion, indicating that SPARC promoted collagen production directly through ADAMTS1 interaction. In conclusion, ADAMTS1 is an important mediator of SPARC-regulated cardiac aging. PMID: 27143554
  7. increased expression of ADAMTS1 protein in macrophages and neutrophils that infiltrated aortic tissues may promote the progression of acute aortic dissection PMID: 26563155
  8. The resilience of casp1(-/-) mice to SIRS lethality is associated with a lower inflammatory response, loss of hippocampal gene coexpression patterns, and increased hippocampal Adamts1 gene expression. PMID: 25633245
  9. Adamts-1 upregulation by inducers of pathological vascular remodeling is mediated by specific signal transduction pathways involving NFAT or C/EBPbeta transcription factors. PMID: 26217013
  10. Western blot analyses indicated that aggrecanase-generated proteoglycan fragments are produced after SCI. PMID: 23562508
  11. Adamts1 potentially contributes to the increased serum levels of CTX in rickets/osteomalacia. PMID: 23261447

  13. ADAMTS1 is associated with regulating neural plasticity. PMID: 23071766
  14. These results reveal that Adamts1 has an impact on bone mineral density and thus further confirm Adamts1 as a potent regulator of bone remodeling. PMID: 22002813
  15. fibulin-1 regulates versican-dependent events in ventricular morphogenesis by promoting ADAMTS1 cleavage of versican leading to suppression of trabecular cardiomyocyte proliferation mediated by the ErbB2-Map kinase pathway. PMID: 22183742
