The DNA fragment encoding the region (Gln20-Gly349) of the mouse GFRAL was expressed with an N-terminal 10xHis-tag and a C-terminal Myc-tag in mammalian cells. The resultant product is the recombinant mouse GFRAL protein. This GFRAL protein has been validated to be active by binding to the mouse GDF15 in a functional ELISA, with the EC50 of 7.926-10.52 ng/mL. Its purity reaches up to 95% measured by SDS-PAGE. It migrated to the molecular weight band of about 45 kDa on the gel. The glycosylation makes the apparent molecular mass slightly higher than the theoretical one. Its endotoxin content is less than 1.0 EU/ug determined by the LAL method. This GFRAL protein is in stock now.

GFRAL is the exclusive receptor for GDF15 which regulates food intake, energy expenditure, and body weight in response to metabolic and toxin-induced stresses. Studies have found that GFRAL is expressed in the posterior brain region of the mouse central nervous system. The GDF15/GFRAL/RET signaling pathway provides new strategies and therapeutic targets for the treatment of metabolism-related diseases, such as obesity, type II diabetes, and pathological overnutrition and malnutrition.
