GDNF-family receptor α-like (GFRAL) belongs to the glial cell-derived neurotrophic factor (GDNF) α-like receptors family. GFRAL is a type I transmembrane protein with a molecular weight of approximately 44 kDa. GFRAL protein is anchored to the cell membrane by glycosylphosphatidylinositol (GPI). As a single-pass transmembrane protein, GFRAL has a short intracellular domain and lacks the ability to conduct signals into cells. Existing studies have found that GFRAL-specific hydrophobic pockets mediate the interaction between GFRAL and the specific ligand GDF15. After binding to GDF15, GFRAL forms a complex with the coreceptor RET, thereby transmitting different downstream signals, which play various biological roles.
In the early days, the in-depth study of GDF-15's biological function was limited due to the lack of its defined receptor. GDF15 is a member of the TGF-β family, but GDF15 does not directly bind to any known TGF-β receptors. Through the screening of human cell membrane-related proteins, GFRAL was finally identified as the only receptor for GDF15. The two have high affinity and can specifically bind to mediate the biological effects of controlling energy intake and energy metabolism regulation. The discovery of the GDF15/GFRAL signaling pathway provides new ideas for the treatment of obesity and metabolic-related diseases, and can be a potential therapeutic target for the treatment of metabolic-related diseases, such as obesity and type II diabetes.
