Description

CUSABIO synthesized the recombinant gene by integrating the N-terminal 6xHis-SUMO tag sequence into the targeted gene encoding the 22-159aa of the mouse Jchain. The synthesized gene was subsequently cloned into an expression vector. After cloning, the expression vector was introduced into the E.coli for expression. The product was purified to obtain the recombinant mouse Jchain protein carrying N-terminal 6xHis-SUMO tag. The SDS-PAGE assayed the purity of this recombinant Jchain protein greater than 90%. This Jchain protein migrated along the gel to a band of about 30 kDa molecular weight.

Immunoglobulin J chain is a protein encoding by a gene named Jchain in mouse and a gene named JCHAIN in human. This protein is a protein component of the antibodies IgM and IgA. The J chain-joined dimer is a nucleating unit for the IgM pentamer. And it induces dimers and/or larger polymers in the case of IgA. Moreover, it also helps to bind these immunoglobulins to secretory component. The diseases involved JCHAIN include Alpha Chain Disease and Climatic Droplet Keratopathy. Additionally, a study have reported that the upregulation of JCHAIN was likely related to tumor aggression, as studied in a cohort of patients with acute lymphoblastic leukemia (ALL).

Target Background

Function
Serves to link two monomer units of either IgM or IgA. In the case of IgM, the J chain-joined dimer is a nucleating unit for the IgM pentamer, and in the case of IgA it induces dimers and/or larger polymers. It also helps to bind these immunoglobulins to secretory component.
Gene References into Functions
  1. Data show that Crlz1 increased the IgJ enhancer activity due to its CBF sites, as verified by both the absence of Crlz1 effect on the CBF-site mutated IgJ enhancer and the presence of transcriptional synergy between Crlz1 and CBFbeta. PMID: 19735751
  2. identifies the HSS3/4 enhancer of Crlz1-IgJ locus as another target of EBG in early developmental stage of B cells PMID: 16962668
  3. H3 and H4 histones toward the chromatin of the Crlz1 gene are found to be hyperacetylated, especially on H3, in the pre-B cells, whereas those toward the chromatin of the IgJ gene are found to be hyperacetylated in the plasma cells PMID: 17015728
