JCHAIN

The Immunoglobulin J chain (encoded by the JCHAIN gene, alias IGJ) is a 15-kDa polypeptide critical for assembling and secreting polymeric immunoglobulins (pIgs), including dimeric IgA and pentameric IgM. It forms disulfide bonds with heavy chains of these pIgs, stabilizing their structure. This modification enables pIgs to bind the polymeric immunoglobulin receptor (pIgR) on mucosal epithelia, facilitating transcytosis into secretions and mediating mucosal immunity.
J chain deficiency reduces secretory IgA/IgM levels, increasing susceptibility to recurrent mucosal infections (e.g., gastrointestinal pathogens). Direct therapeutic targeting is emerging: strategies to upregulate J chain in B cells aim to restore mucosal immunity in immunodeficiencies. Aberrant J chain expression in B cell malignancies (e.g., multiple myeloma) also suggests its potential as a diagnostic biomarker.
