Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The production of this recombinant Mouse Il31ra protein is just like all recombinant proteins. The process involved transfecting E.coli cells with DNA vector containing the template of recombinant DNA. The E.coli cells containing the template were then cultured so that they could transcribe and translate the Il31ra protein. N-terminal 6xHis tag was used in the process. The purity is 0.85 determined by SDS-PAGE.

The receptor IL-31RA is the receptor of IL-31, which has been found unregulated in skin and lung fibroblasts from Scleroderma. IL-31 acts via a heterodimeric receptor comprising IL-31RA and oncostatin M, expressed by multiple cell types, and signalling via PI3K, ERK and JAK/STAT pathways. Interleukin (IL)-31 is a cytokine produced mainly by Th2 cells. And it signals via the IL-31RA and oncostatin M receptor β heterodimer. IL-31RA is expressed in both human skin and dorsal root ganglions. IL-31RA is a critical neuroimmune link between Th2 cells and the sensory nerves. Some resports suggested that that IL-31RA is expressed not only in nerve fibres but also in keratinocytes in the dermis from Alzheimer patients. Besides, researchers observed that administration of IL-31 upregulates IL-31RA in dorsal root ganglia and causes severe itch-associated scratching behaviour in mice.

Target Background

Function
Associates with OSMR to form the interleukin-31 receptor which activates STAT3 and to a lower extent STAT1 and STAT5. May function in skin immunity. Mediates IL31-induced itch, probably in a manner dependent on cation channels TRPA1 and TRPV1. Positively regulates numbers and cycling status of immature subsets of myeloid progenitor cells in bone marrow in vivo and enhances myeloid progenitor cell survival in vitro.
Gene References into Functions
  1. Anti-IL-31 receptor antibody is shown to be a potential therapeutic option for treating itch and dermatitis in mice PMID: 24946165
  2. IL31RA is a functional receptor expressed by a small subpopulation of IL-31RA(+)/TRPV1(+)/TRPA1(+) neurons and is a critical neuroimmune link between TH2 cells and sensory nerves for the generation of T cell-mediated itch. PMID: 24373353
  3. Gp130(757F/F) mice rapidly develop distal stomach tumors, with loss of SHP2/Erk/AP-1 transcriptional regulation exemplified by decreased TFF1 expression and increased STAT1/3 regulated genes such as Reg I. PMID: 14699500
  4. IL-31 receptor alpha signaling is a novel negative regulatory pathway that specifically limits T helper cell type 2 inflammation. PMID: 17353366
Subcellular Location
Cell membrane; Single-pass type I membrane protein. Cell junction, synapse, presynaptic cell membrane. Cell projection, axon.
Protein Families
Type I cytokine receptor family, Type 2 subfamily
Tissue Specificity
Expressed in a subset of dorsal root ganglia neurons. Expressed in spinal cord and trigeminal ganglion (at protein level). Expressed in skin, testis, bone marrow and thymus.
