Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Is a key player in the control of plasma membrane curvature, and membrane shaping and remodeling. Required in muscle cells for the formation of T-tubules, tubular invaginations of the plasma membrane that function in depolarization-contraction coupling. Required in muscle cells for the formation of T-tubules, tubular invaginations of the plasma membrane that function in depolarization-contraction coupling. Is a negative regulator of endocytosis. Is also involved in the regulation of intracellular vesicles sorting, modulation of BACE1 trafficking and the control of amyloid-beta production. In neuronal circuits, endocytosis regulation may influence the internalization of PHF-tau aggregates. May be involved in the regulation of MYC activity and the control cell proliferation.
Gene References into Functions
  1. Data (including data from studies using transgenic mice) suggest that the process leading to microparticle release from cardiac myocytes involves recruitment of CHMP4B protein to the forming microparticle membrane which also contains cBIN1; plasma cBIN1 is reduced in patients with heart failure as compared to control subjects. (CHMP4B = charged multivesicular body protein 4B; cBIN1 = cardiac bridging integrator 1) PMID: 28806752
  2. Bin1 and CD2AP keep APP and BACE1 apart in early endosomes by distinct mechanisms in axon and dendrites. Individuals carrying variants of either factor would slowly accumulate Abeta in neurons increasing the risk for late-onset AD. PMID: 27895104
  3. Data demonstrate that EHBP1L1 links Rab8 and the Bin1-dynamin complex, which generates membrane curvature and excises the vesicle at the endocytic recycling compartment for apical transport. PMID: 26833786
  4. Findings show how cardiac deficiency in Bin1 function causes age- and stress-associated heart failure suggesting that Bin1 is a positive modifier of cardiac contractility that helps sustain adult heart function under stress conditions. PMID: 25939245
  5. Reorganization of BIN1-induced microdomains recruits phosphorylated ryanodine receptors into dyads, increasing calcium signaling. PMID: 26733606
  6. Bin1 mAb reduced colitis morbidity in mice while unprotected mice were characterized by severe lesions throughout the mucosa, rupture of lymphoid follicle, high-level neutrophil and lymphocyte infiltration into the mucosal areas, loss of surface crypts. PMID: 26195312
  7. The release of BIN1 from hypo-poly(ADP-ribosyl)ated E2F1 is a mechanism by which serum starvation promotes E2F1-induced apoptosis. PMID: 25257171
  8. BIN1 interacts with MTM1 in skeletal muscle. PMID: 23917616
  9. Cardiac BIN1 folds T-tubule membrane, controlling ion flux and limiting arrhythmia. PMID: 24836577
  10. Bin1 is a genetic modifier of colitis that controls the paracellular pathway of transcellular ion transport regulated by cellular tight junctions. PMID: 22526583
  11. Data have identified that membrane-associated BIN1 not only induces membrane curvature but can direct specific antegrade delivery of microtubule-transported membrane proteins. PMID: 20169111
  12. BIN1 is transcriptionally activated by E2F1 and mediates E2F1-induced apoptosis in response to DNA damage. PMID: 19629135
  13. findings support a role of the bilayer-deforming properties of amphiphysin at T-tubules and, more generally, a physiological role of amphiphysin in membrane deformation PMID: 12183633
  14. Sorting nexin 4 and amphiphysin 2 have roles in endocytosis and intracellular trafficking PMID: 12668730
  15. Bin1 has a developmentally critical role in cardiac muscle development PMID: 12773571
  16. the results define a function for Bin1 in apoptosis that is conditional on transformation stress. PMID: 12789266
  17. findings suggest that Bin1 loss promotes immune escape in cancer by deregulating indoleamine 2,3-dioxygenase (IDO) and that IDO inhibitors may improve responses to cancer chemotherapy PMID: 15711557
  18. In mouse tumors, Bin1 ablation facilitated progression to more aggressive invasive status. PMID: 17699764
  19. Bin1 displays protein scaffold-like properties and binds with sarcomeric factors important in directing sarcomere protein assembly and myofiber maturation. PMID: 19633357
  20. Bin1 deletion elevated female reproductive capability until the age of 17.3 +/- 1.1 months and increased the incidence of lung cancer, liver cancer, and premalignant lesions in the prostate during aging. PMID: 17699764
  21. Mammary specific deletion of Bin1 cooperates with carcinogen-induced ras activation to drive mammary tumor progression, establishing a role for Bin1 as a negative modifier of oncogenicity and progression in breast cancer. PMID: 17210688
  22. Bin1 is upregulated in somites during muscle development; Bin1 induction in differentiation is required for p21WAF1 upregulation. PMID: 10036185
