Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Possibly involved in cell-cell and cell-matrix interactions during inflammation and tumorigenesis.
Gene References into Functions
  1. TSG-6 deficiency protects against airway hyperresponsiveness after ozone (in vivo) or short-fragment hyaluronan (sHA) (in vitro and in vivo) exposure. Moreover, TSG-6(-/-) tracheal ring non-responsiveness to sHA was reversed by exogenous TSG-6 addition. PMID: 29122888
  2. Loss of TSG6 expression is associated with Embryos failing to Initiate Midgut Rotation. PMID: 30174180
  3. TSG-6 up-regulation counteracts the progression of atherosclerosis. (Review) PMID: 29401724
  4. our results showed that the increase of oxidative stress, activation of the NLRP3 inflammasome and NF-kappaB signaling in SAP was substantially inhibited following administration of hMSCs or TSG-6, which was dependent on the presence of CD-44 receptors in acinar cells. PMID: 27917949
  5. TSG-6 protects hepatocytes from damage by enhancing autophagy influx and contributes to liver regeneration PMID: 28935975
  6. TSG6 functions by converting macrophages from a proinflammatory to an anti-inflammatory phenotype secondary to suppression of TLR4/NF-kappaB signaling and STAT1 and STAT3 activation. PMID: 27911817
  7. This study provided the first evidence of TSG-6 secreted by mesenchymal stem cells promoting corneal epithelial wound healing in diabetic mice through activating corneal epithelial stem/progenitor cells and accelerating M2 macrophage polarization PMID: 28810264
  8. This study shows that TSG-6 is expressed in the CNS, suggesting a role for TSG-6 in astrocyte activation and tissue repair. PMID: 27435674
  9. Data (including data from studies in knockout mice) suggest that TSG-6 is needed for release of hyaluronan rafts (leukocyte-adhesive rafts) from apical surface of tracheal mucosa into bronchoalveolar lavage fluid as heavy chain hyaluronan complexes. PMID: 26601955
  10. the well characterized hyaluronan-binding site in the TSG-6 Link module is not used for recognition during transfer of HCs onto HA. PMID: 26468290
  11. Thus, TSG6, HA, and IalphaI were crucial factors for the settlement and probably the subsequent differentiation of MSCs. PMID: 26178374
  12. BM-MSCs injected into mice with colitis do not localize to the intestine but instead form aggregates in the peritoneum where they produce immunoregulatory molecules, including TSG6, that reduce intestinal inflammation. PMID: 25790743
  13. This study demonstrated that following stimulation with TNF-alpha, MSCs modulate microglia activity through TSG-6. PMID: 25088370
  14. suggest that TSG-6 plays an important role in MSCs-mediated immunosuppressive effect on DC PMID: 25014173
  15. This study provides insight into what we believe to be a previously undescribed multifaceted role of mesenchymal stem cell-released TSG-6 in wound healing. PMID: 23921952
  16. TSG-6 is implicated in the experimental murine model of allergic pulmonary inflammation and is likely to contribute to the pathogenesis of asthma PMID: 23118230
  17. TSG-6 increases the accumulation of HA in the cell-associated matrix, partially by preventing its dissolution from the cell-associated matrix into the conditioned medium, but primarily by inducing HA synthesis PMID: 23129777
  18. analysis of irreversible heavy chain transfer to hyaluronan oligosaccharides by tumor necrosis factor-stimulated gene-6 PMID: 23166324
  19. cells expressing CD44/TSG-6 failed to roll on hyaluronan, instead, they stuck and remained "tethered" to the substrate under fluid flow PMID: 12011075
  20. observations identify tumor necrosis factor-alpha-stimulated gene 6 as a target of prostaglandin action and show that its production in ovulatory follicles is associated with proper formation of the cumulus-derived extracellular matrix PMID: 12586778
  21. results provide clear evidence that TNFIP6 is a key catalyst in the formation of the cumulus extracellular matrix and indispensable for female fertility PMID: 12668637
  22. TSG-6 mRNA is induced in cumulus cells in culture by cAMP and that the secreted TSG-6 protein is a key structural component of the mouse cumulus cell-oocyte complex matrix. PMID: 12959984
  23. Tnsf6 plays the dominant role in both WT (WildType) and KO (connexin43-null) mice heart. PMID: 14756057
  24. The ability of TSG-6 to modulate the interaction of hyaluronan with CD44 has important implications for CD44-mediated cell activity at sites of inflammation, where TSG-6 is expressed. PMID: 15060082
  25. TSG-6 protein forms stable complexes with hyaluronan and binds to chondroitin sulfates PMID: 15699048
  26. The tumor necrosis factor alpha induced protein 6 is as a potential marker that could be used for early diagnosis and prognosis of cancerous or precancerous lesions. PMID: 16467113
