Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

The recombinant Naja kaouthia Alpha-CT protein is encoded by the gene of Alpha-CT (1-71aa). The gene of Alpha-CT was cloned in a system (E.coli) that supported the expression of Alpha-CT. Modification of Alpha-CT by recombinant DNA technology could lead to the expression of the target protein. The protein was fused with N-terminal 10xHis-GST tag & C-terminal Myc tag in the production. The purity is 85% determined by SDS-PAGE.

Alpha-cobratoxin is a nicotinic acetylcholine receptor. According to some research, Alpha-cobratoxin may have the following features.
Alpha-cobratoxin as a possible treatment for multiple sclerosis. Alpha-cobratoxin inhibits the T-type calcium current of dorsal root ganglion neurons through the muscarinic M4 receptor and Go-protein βγ subunit-dependent protein kinase A pathway. Isolation and characterization of high-affinity VHH antibody fragments against alpha-cobratoxin.

Monomer: binds with high affinity to muscular (alpha-1-beta-1-gamma-delta/CHRNA1-CHRNB1-CHRNG-CHRND) nAChR (tested on Torpedo californica, Kd=0.2-4.5 nM) and neuronal alpha-7/CHRNA7 nicotinic acetylcholine receptors (Kd=13-105 nM). Also inhibits GABA(A) channels. Heteropentamer targets studied are composed of alpha-1-beta-3-gamma-2 (GABRA1-GABRB3-GABRG2) subunits (IC(50)=236 nM), alpha-1-beta-2-gamma-2 (GABRA1-GABRB2-GABRG2) subunits (IC(50)=469 nM), alpha-2-beta-2-gamma-2 (GABRA2-GABRB2-GABRG2) subunits (IC(50)=485 nM), alpha-5-beta-3-gamma-2 (GABRA5-GABRB3-GABRG2) subunits (IC(50)=635 nM), and alpha-2-beta-3-gamma-2 (GABRA2-GABRB3-GABRG2) subunits (IC(50)=1099 nM) (activated by 10 uM GABA).; Homodimer: binds with high affinity (but lower than the monomeric form) to muscular (IC(50)=9.7 nM) and with low affinity to neuronal alpha-7/CHRNA7 nAChRs (IC(50)=1370 nM).
