Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

Our Recombinant Pseudomonas aeruginosa lasB is a bioengineered replica of the native Elastase protein. The lasB gene, sourced from Pseudomonas aeruginosa strain ATCC 15692 / PAO1 / 1C / PRS 101 / LMG 12228, was expressed in E.coli and covers the 198-498 amino acid region, which represents the full length of the mature protein.

The resulting product carries a N-terminal 6xHis-SUMO-tag to facilitate detection and purification processes. After extraction, our Recombinant Pseudomonas aeruginosa lasB undergoes rigorous testing to ensure its purity is greater than 90%, as determined by SDS-PAGE. We provide this product in either liquid or lyophilized powder form to accommodate different research needs and experimental setups.

 Q&A
Q:

Item CSB-EP318584EZX on PO 41807 is now in Tris-HCl buffer with EDTA. We received this product before and it was in PBS. Is this a permanent product change?

A:
Thanks for your inquiry,Yes, if there is no special request, we will use 10 mM Tris-HCl,1 mM EDTA,pH8.0 as the buffer. If you still need PBS buffer, pls write on the PO. We will process like that.

Target Background

Function
Cleaves host elastin, collagen, IgG, and several complement components as well as endogenous pro-aminopeptidase. Autocatalyses processing of its pro-peptide. Processes the pro-peptide of pro-chitin-binding protein (cbpD). Involved in the pathogenesis of P.aeruginosa infections.
Gene References into Functions
  1. This study represents a systematic mutational analyses of salt bridges in the model metalloprotease PAE and provides important insights into the structure-function relationship of enzymes PMID: 25815820
  2. both bactericidal and cell-recruiting activities of MIG/CXCL9 are corrupted by P. aeruginosa through release of elastase, and this may contribute to impaired airway host defense in CF. PMID: 25115612
  3. Data indicate that the purified elastase protein had exhibited a homodimer size of 65 kDa by SDS-PAGE and MALDI-TOF, a size which is totally distinct from that of typically reported 33 kDa monomer. PMID: 22016627
  4. the protease LasB, directly affected VE-cadherin proteolysis PMID: 24626230
  5. P. aeruginosa elastase transiently disrupts tight junctions in human nasal epithelial cells and downregulates PAR-2. PMID: 24548792
  6. Elastase cleaves corticosteroid-binding globulin reactive center loop at a site distinct from that targeted by neutrophil elastase. PMID: 24848868
  7. Gene expression analysis revealed that the mutant of lasB could result in the downregulation of rhamnolipid synthesis at the transcriptional level. PMID: 24693981
  8. Among Pseudomonas aeruginosa-secreted metalloproteinases, LasB can induce vascular cell anoikis through simultaneous proteolysis of ECM components and cell receptors, suggesting the uPAR-vitronectin axis as a major target in this process. PMID: 21501369
  9. LasB is able to cleave uPAR both within the sequence linking D1 to D2 and at the carboxy terminus of D3. PMID: 17517866
