Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Microtubule-associated force-producing protein involved in producing microtubule bundles and able to bind and hydrolyze GTP. Most probably involved in vesicular trafficking processes. Involved in receptor-mediated endocytosis.
Gene References into Functions
  1. Dynamin was identified as a septin-binding protein within neuronal membrane raft. PMID: 23260429
  2. Compared to endophilinI/II, the interaction between dynamin I and endophilin III is significantly increased in clathrin-mediated endocytosis. PMID: 22961472
  3. Phosphorylation and nitration of tyrosine residues affect functional properties of Synaptophysin and Dynamin I. PMID: 23103755
  4. EHD1 inhibits dynamin-induced membrane tubulation in the presence of GTPgammaS, a phenomenon linked with dynamin helix assembly in synaptic vesicles. PMID: 21957258
  5. Ca(2+) influx regulates assembly of a fully active CaN-calmodulin complex selectively on the tail of dynIxb and the complex is recruited to sites of activity-dependent bulk endocytosis in nerve terminals PMID: 21730063
  6. In a rat model of Alzheimer's disease, a decrease of dynamin I in hippocampal neurons is found at protein and mRNA levels. PMID: 20847448
  7. The deformation dynamics of dynamin is dominated by the draining of the membrane out of the helix, allowing quantification of helix-membrane interactions. PMID: 21112282
  8. Study found that the specific phosphorylation of Ser-774 on dynamin I by glycogen synthase kinase 3 was both necessary and sufficient for activity-dependent bulk endocytosis. PMID: 20526333
  9. These findings suggest that evoked synchronous and asynchronous release originate from the same vesicle pool that recycles rapidly in a dynamin-dependent manner; a distinct vesicle pool sustains spontaneous release independent of dynamin activation. PMID: 20107062
  10. developed a theory based on the competition between dynamin polymerization and membrane mechanical deformation. This curvature control of dynamin polymerization is predicted for a specific range of concentrations, which corresponds to our measurements PMID: 20160074
  11. couples endocytosis and exocytosis PMID: 11856729
  12. Mnbk/Dyrk1A plays a dual role in regulating the interaction of dynamin 1 with amphiphysin 1. Phosphorylation reduced the interaction of dynamin with endophilin 1,the same phosphorylation enhanced the binding of dynamin 1 to Grb2; important to endocytosis. PMID: 11877424
  13. demonstrate that phosphorylation of dynamin I by c-Src induces its self-assembly and increases its GTPase activity PMID: 12011079
  14. Results of dynamin 1 mutants study offer direct evidence that supports the conclusion that fast site Ser-857 phosphorylation causes reduction in the level of binding of dynamin 1 to amphiphysin 1 and directly contributes to enhanced Grb 2 binding. PMID: 15287745
  15. Interaction with phospholipase D2 may be involved in enzyme activation. PMID: 15581494
  16. Dynamin 1 is a novel dystroglycan-associated protein. PMID: 15728588
  17. results support a model in which switch I residue Arg-59 gates GTP binding in an assembly-dependent manner and the GTPase effector domain functions as an assembly-dependent GTPase activating protein in the fashion of RGS-type GAPs. PMID: 16141317
  18. dynamin 1 degradation was the result of calpain activation induced by the sustained calcium influx mediated by N-methyl-D-aspartate receptors in hippocampal neurons. PMID: 16864575
  19. Because syndapin acts as a phospho-sensor, this supports its role in depolarization-induced SVE at the synapse, which involves dynamin dephosphorylation. PMID: 17437541
  20. Amyloid-beta caused a disruption of synaptic vesicle endocytosis in cultured hippocampal neurons; evidence suggests Abeta-induced dynamin 1 depletion plays a role in this process. PMID: 17499934
  21. SNX9 interacts with dynamin-1 and N-WASP in presynaptic terminals, where it links actin dynamics and synaptic vesicle endocytosis. PMID: 17681954
  22. The tandem SH3A and SH3B domains of Tks5 constitute a versatile module for the implementation of isoform-specific protein-protein interactions. PMID: 19464300
