Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

To achieve the expression of the recombinant Vaccinia virus (strain Western Reserve) VACWR156 protein in Yeast cells, a DNA fragment encoding the Vaccinia virus (strain Western Reserve) VACWR156 protein (57-185aa) is inserted into a plasmid vector, which is then transferred to Yeast cells. After screening positive cells, they are cultured and induced to generate the VACWR156 protein. A N-terminal 6xHis tag is attached to the protein. Cells are lysed to collect the recombinant Vaccinia virus (strain Western Reserve) VACWR156 protein, which is purified through affinity purification and then identified using SDS-PAGE and subsequent staining of the gel with Coomassie Brilliant Blue. The purity of the obtained recombinant Vaccinia virus (strain Western Reserve) VACWR156 protein is over 85%.

Target Background

Function
Coordinates the incorporation of A36 into wrapped enveloped virion (EV) membranes and, subsequently, the production of actin tails. Therefore plays an essential role in efficient cell-to-cell spread of viral particles.
Gene References into Functions
  1. The localization and stability of A34 is affected by B5 and here data are presented showing that A34 is also affected by A33. PMID: 23255618
  2. The authors report that A33 is present as a disulfide-bonded homodimer during infection. PMID: 20947114
  3. Comparison of the three-dimensional A33 model to the X-ray structures of distant cellular homologues revealed that A33 retained the key residues required for adopting the C-type lectin-like fold. PMID: 20302896
  4. findings show that vaccinia virus spreads across one cell every 75 minutes; early expression of proteins A33 and A36 was critical for virion repulsion and rapid spread, and cells expressing these proteins repelled exogenous virions rapidly PMID: 20093437
Subcellular Location
Virion membrane; Single-pass type II membrane protein. Host membrane; Single-pass type II membrane protein.
Protein Families
Chordopoxvirinae A33 protein family
