  • Overlay Peak curve showing HT29 cells stained with CSB-RA188971A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

MUC2 serves as the principal gel-forming mucin in the intestinal tract, where it establishes the protective mucus barrier that separates luminal contents from the underlying epithelium. This secreted glycoprotein plays a fundamental role in maintaining intestinal homeostasis, and its dysregulation has been implicated in inflammatory bowel diseases, colorectal cancer progression, and mucinous tumor phenotypes. Understanding MUC2 expression patterns and regulation provides critical insights into gastrointestinal pathophysiology and mucosal immunity.

This recombinant monoclonal antibody, clone 14H6, offers the reproducibility and consistency that demanding research applications require. Generated against a synthetic peptide derived from human MUC2 and produced using recombinant technology in rabbit host, this antibody delivers sequence-defined specificity with minimal lot-to-lot variation. The monoclonal nature ensures that every experiment draws from the same defined binding characteristics, supporting reliable data across longitudinal studies and multi-site collaborations.

Validation in flow cytometry demonstrates clear detection of MUC2 in HT29 cells, a well-established human colorectal adenocarcinoma line known for mucin production. The intracellular staining protocol, employing fixation and permeabilization to access this secreted protein, showed distinct signal separation from isotype control, confirming specific target recognition. Recommended dilutions for flow cytometry range from 1:50 to 1:200, providing flexibility to optimize signal intensity for your specific experimental conditions. The antibody is also validated for ELISA applications.

Supplied in a stabilized liquid format with glycerol for convenient long-term storage at -20°C or -80°C, this antibody supports researchers investigating mucosal biology, gastrointestinal cancers, inflammatory conditions, and epithelial barrier function.
