Description

NTRK1, also known as TrkA, serves as the high-affinity receptor for nerve growth factor and plays a fundamental role in neuronal development, survival, and differentiation. As a receptor tyrosine kinase, TrkA signaling influences pain perception, neurotrophin-mediated processes, and has emerged as a significant focus in oncology research due to NTRK gene fusions identified across multiple tumor types.

This recombinant monoclonal antibody, clone 2A5, offers the reproducibility and consistency that demanding research protocols require. Because the antibody sequence is defined and produced recombinantly in rabbit host cells, you can expect uniform performance across experiments and over time, eliminating the lot-to-lot variability that can complicate long-term studies or multi-site collaborations.

Validated for Western blot applications at dilutions ranging from 1:500 to 1:5000, this antibody has demonstrated reliable detection in rat brain and mouse brain whole cell lysates. The observed band at approximately 145 kDa, which differs from the predicted molecular weights of 78-88 kDa, reflects the extensive post-translational modifications characteristic of TrkA, including glycosylation that substantially increases the receptor's apparent molecular weight on SDS-PAGE. This detection pattern is consistent with mature, fully processed TrkA protein. The antibody provides cross-species reactivity across human, mouse, and rat samples, offering flexibility for researchers working with different model systems or translational studies.

Receptor tyrosine kinase involved in the development and the maturation of the central and peripheral nervous systems through regulation of proliferation, differentiation and survival of sympathetic and nervous neurons. High affinity receptor for NGF which is its primary ligand. Can also bind and be activated by NTF3/neurotrophin-3. However, NTF3 only supports axonal extension through NTRK1 but has no effect on neuron survival. Upon dimeric NGF ligand-binding, undergoes homodimerization, autophosphorylation and activation. Recruits, phosphorylates and/or activates several downstream effectors including SHC1, FRS2, SH2B1, SH2B2 and PLCG1 that regulate distinct overlapping signaling cascades driving cell survival and differentiation. In absence of ligand and activation, may promote cell death, making the survival of neurons dependent on trophic factors.; Resistant to NGF, it constitutively activates AKT1 and NF-kappa-B and is unable to activate the Ras-MAPK signaling cascade. Antagonizes the anti-proliferative NGF-NTRK1 signaling that promotes neuronal precursors differentiation. Isoform TrkA-III promotes angiogenesis and has oncogenic activity when overexpressed.
