Titin stands as the largest known protein in the human body, functioning as a molecular spring that spans half the sarcomere from the Z-disc to the M-line. This giant protein plays essential roles in muscle elasticity, sarcomere assembly, and mechanosensing, making it a critical target for researchers investigating cardiomyopathies, muscular dystrophies, and skeletal muscle disorders. Mutations in the TTN gene are among the most common genetic causes of dilated cardiomyopathy, while also being implicated in limb-girdle muscular dystrophy type 2J and hereditary myopathy with early respiratory failure.
This recombinant monoclonal antibody, clone 12D11, offers the consistency and reliability that titin research demands. Developed using recombinant technology with a defined human IgG1 isotype, this antibody provides sequence-defined specificity and eliminates the lot-to-lot variability that can complicate longitudinal studies of this complex protein. The antibody was raised against recombinant human TTN protein and purified by affinity chromatography, ensuring high purity for demanding applications.
Validation through immunohistochemistry demonstrates clear detection in paraffin-embedded human tissues, with successful staining confirmed in both skeletal muscle and liver cancer tissue sections at dilutions of 1:50. These results, obtained using citrate buffer antigen retrieval on a Leica Bond system, provide researchers with optimized starting conditions for their own investigations. The antibody is also validated for ELISA applications, offering flexibility across experimental platforms.
This TTN antibody serves as a valuable tool for cardiovascular researchers studying inherited cardiomyopathies, muscle biologists investigating sarcomere structure and function, and pathologists examining muscle-related malignancies.
