Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Glycoprotein that probably modulates membrane fusion events during secondary envelopment of cytoplasmic capsids that bud into specific trans-Golgi network (TGN)-derived membranes. Also plays a role, together with gB, in virus-induced cell-to-cell fusion (syncytia formation). Seems to block fusion of virions with infected-cell membranes.
Gene References into Functions
  1. a deletion in the amino terminus of glycoprotein K (gK) inhibits gB binding to Akt-1(S473), the release of intracellular calcium, and virus entry via fusion of the viral envelope with cellular plasma membranes. PMID: 29321326
  2. This work suggests that the amino terminus of gK is a critical determinant for entry into neuronal axons and may serve similar conserved functions for other alphaherpesviruses. PMID: 26656706
  3. Taken together, these results strongly suggest that the 8mer peptide (ITAYGLVL) within the signal sequence of gK promotes cell surface expression of gK in infected cells and ocular pathogenesis in infected mice. PMID: 25505072
  4. We demonstrate a novel link between gK exacerbation of CS and HSV-1 receptors, suggesting a gK-induced molecular route for the pathogenesis as well as selective advantage of these entry routes for the pathogen during latency-reactivation cycle. PMID: 24667863
  5. The amino terminus of herpes simplex virus 1 gK is functionally and physically associated with the gB-PILRalpha protein complex and regulates membrane fusion of the viral envelope with cellular membranes during virus entry. PMID: 23302878
  6. gK is a critical determinant for neuronal infection and transmission. PMID: 22829677
  7. required for gB mediated virus induced cell fusion but does not function redundantly with gK and UL20p in membrane fusion during virion de-envelopment at the outer nuclear lamellae. PMID: 15596825
  8. These results strongly suggest that increased gK levels promote eye disease and chronic infection in infected mice. PMID: 17898051
  9. These results show that UL20p domains required for UL20p and gK intracellular transport and TGN localization can be functionally segregated from domains involved in infectious virus production and virus-induced cell fusion. PMID: 17996071
  10. These data show that the amino terminus of gK modulates gB-mediated virus-induced cell fusion and virion egress. PMID: 19793812

Subcellular Location
Host cell membrane; Multi-pass membrane protein. Host endosome membrane; Multi-pass membrane protein. Host Golgi apparatus membrane; Multi-pass membrane protein. Note=During virion morphogenesis, this protein probably accumulates in the endosomes and trans-Golgi where secondary envelopment occurs. It is probably transported with UL20 to the cell surface from where it is endocytosed and directed to the trans-Golgi network (TGN). Cell surface expression of gK is required for virus-induced cell-to-cell fusion. Likely not present in extracellular virions.
