The process of generating the DBP polyclonal antibody is methodical and precise. It commences with the repeated immunization of a rabbit using a recombinant human adenovirus C serotype 5 DBP protein (174-529aa), continuing until a sufficient antibody titer is achieved. Subsequently, the rabbit's blood is collected, and the antibodies are meticulously purified from the serum utilizing protein A/G. The functionality of the resulting DBP antibody is comprehensively evaluated through ELISA and WB applications, confirming its specific binding to the human adenovirus C serotype 5 DBP protein.
DBP is the major viral replication protein generated during human adenovirus C serotype 5 infection. DBP binds dsDNA and ssDNA, and it changes the DNA structure drastically. DBP is involved in several steps during DNA replication, including protecting ssDNA against nucleases, increasing the rate and processivity of AdV DNA polymerase and changing its sensitivity to inhibitors during elongation, and driving template unwinding by the formation of DBP multimers.
