The development of polyclonal antibodies against ogt requires repetitive immunizations of a rabbit with recombinant Escherichia coli (strain K12) ogt protein (1-171aa). Upon achieving a satisfactory antibody titer, the rabbit undergoes bleeding, and antibodies are purified from the serum via affinity chromatography. The resulting ogt antibody is then evaluated for functionality in ELISA and WB applications, demonstrating its capability to recognize Escherichia coli (strain K12) ogt protein.
The ogt protein in Escherichia coli (strain K12) plays a crucial role in DNA repair. Its main function involves the removal of alkyl groups, such as methyl or ethyl groups, from the O6 position of guanine in DNA. This repair activity is targeted at addressing DNA damage caused by exposure to alkylating agents, both endogenous and exogenous, thereby preserving the integrity of the DNA molecule. By detoxifying alkylating agents and maintaining genomic stability, ogt contributes to the cell's ability to cope with environmental stresses and challenges.
