Description

In the generation of the pum polyclonal antibody, a rabbit is immunized with a recombinant Drosophila melanogaster (Fruit fly) pum protein segment (1091-1330aa) to trigger an antibody response. The rabbit serum is obtained and then purified using affinity chromatography to harvest polyclonal antibodies against pum. The pum antibody is recommended for detecting the Drosophila melanogaster (Fruit fly) pum protein through ELISA and WB assays.

The Pumilio (Pum) protein in Drosophila melanogaster is a crucial regulator of post-transcriptional gene expression. Acting as an RNA-binding protein, Pumilio binds to specific sequence motifs in the 3' untranslated regions of target mRNAs, leading to the repression of translation and mRNA degradation. Pumilio plays pivotal roles in various biological processes, including germline development, where it regulates the expression of genes involved in germ cell fate and differentiation. In embryonic development, Pumilio is essential for establishing polarity and proper patterning by controlling the translation of key mRNAs.

Target Background

Function
Sequence-specific RNA-binding protein that acts as a post-transcriptional repressor by binding the 3'-UTR of mRNA targets. Binds to an RNA consensus sequence, the Pumilio Response Element (PRE), 5'-UGUANAUA-3', that is related to the Nanos Response Element (NRE). Mediates post-transcriptional repression of transcripts via different mechanisms: acts via direct recruitment of deadenylase complexes leading to translational inhibition and mRNA degradation. Also mediates deadenylation-independent repression by promoting accessibility of miRNAs. Mediates post-transcriptional silencing of E2f mRNA by binding to its 3'-UTR and promoting miRNA regulation. Required for abdominal development and to support proliferation and self-renewal of germ cells. Pum is the only gene required for nos activity that is not also required for posterior localization of germline determinants. Pum is required during embryogenesis when nos activity apparently moves anteriorly from the posterior pole.
