Description

In the production of the US12 antibody, a rabbit is immunized with recombinant human herpesvirus 1 ICP47 protein (1-88aa), resulting in the production of IgG antibodies by the rabbit's B lymphocytes. The polyclonal US12 antibody is then subjected to meticulous purification through protein G. This US12 antibody can react with human herpesvirus 1 ICP47 protein in ELISA and WB experiments.

The human herpesvirus 1 (HSV-1) US12-encoding protein ICP47 is a polymorphous protein. HSV-1 ICP47 could interfere with RNA splicing in early infection and then shuttle viral mRNA from the nucleus to the cytoplasm in late infection. HSV-1 ICP47's direct interaction with the antigen-dependent transporter (TAP) restricts antigen trafficking, resulting in the generation of empty MHC-I molecules, thus promoting the viral evasion of the immune system.

Function
Plays a role in the inhibition of host immune response. Binds specifically to transporters associated with antigen processing (TAP), thereby blocking peptide-binding and translocation by TAP as well as subsequent loading of peptides onto MHC class I molecules. Empty MHC I molecules are retained in the endoplasmic reticulum and ultimately directed to proteasomal degradation. In consequence, infected cells are masked for immune recognition by cytotoxic T-lymphocytes.
