To create the psaA polyclonal antibody, a recombinant Streptococcus pneumoniae psaA protein fragment (comprising amino acids 21 to 310) serves as the chosen immunogen. This fragment is introduced into a rabbit for immunization, prompting the development of an antibody response. Subsequently, serum is obtained from the rabbit and purified through affinity chromatography to obtain polyclonal antibodies. Rigorous validation is conducted to confirm the psaA antibody's specificity for Streptococcus pneumoniae psaA protein through ELISA and WB assays.
Pneumococcal surface adhesin A (psaA) is a lipoprotein component of the Mn2+ transporter, found on the surface of all known serotypes of Streptococcus pneumoniae. It is an adhesin that plays a crucial role in the attachment of pneumococci to host cells and contributes to their virulence. psaA is also immunogenic and natural colonization of the nasopharynx by pneumococci leads to an increase in antibody production against PsaA.
