The product CSB-EL011616HU is a sandwich ELISA kit developed to measure levels of human interleukin-36 receptor antagonist protein (IL-36Ra) in serum, plasma, or tissue homogenates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of IL1F5 bound in the initial step.
IL-36Ra, also called IL1F5, functions as a receptor antagonist that inhibits the activation of IL-36R signaling. It induces inflammatory responses by activating NF-κB and MAPKs via an intracellular signaling cascade. In contrast, IL-36Ra suppresses pro-inflammatory signaling by binding to and inhibiting IL-36R activation. It is highly expressed in the skin, brain, tonsils, spleen, and various immune cell types such as macrophages, monocytes, B cells, and dendritic cells (DCs). Loss-of-function homozygous or compound heterozygous mutations in its encoding gene IL36RN have been implicated in the pathogenesis of various skin disorders.
