IL-36 receptor antagonist (IL-36Ra) plays a critical role in dampening IL-36-driven inflammatory signaling, making it an essential tool for dissecting NF-κB and MAPK pathway regulation in innate immunity. This tag-free recombinant human IL-36RN encompasses the full-length mature sequence (residues 2–155), expressed in *E. coli* and confirmed fully active with an ED50 below 1.0 μg/ml in an IL-36β-induced IL-8 secretion assay using human preadipocytes, corresponding to a specific activity exceeding 1,000 IU/mg. Endotoxin levels below 1.0 EU/μg minimize LPS-driven artifacts, which supports use in sensitive cell-based functional assays such as immune cell activation and differentiation studies, as well as in vivo inflammation models where confounding innate immune stimulation must be controlled. Purity exceeding 95% by SDS-PAGE, combined with this endotoxin profile, provides a suitable basis for applications including antibody validation, ELISA standard curves, and cytokine detection reference standards.
