The product CSB-E06873h is a sandwich ELISA kit developed to measure concentrations of human ACTH in serum, plasma, or tissue homogenates. This assay uses the sandwich enzyme immunoassay technique in combination with the enzyme-substrate chromogenic reaction to quantify the analyte in the sample. The color develops positively to the amount of ACTH in samples. The color intensity is measured at 450 nm via a microplate reader.
ACTH is an amino acid peptide hormone produced by cells of the anterior pituitary gland and carried by the peripheral circulation to the adrenal cortex, where it stimulates the synthesis and secretion of glucocorticoids and, to a more modest extent, mineralocorticoids and adrenal androgens. As the anterior pituitary mediator of the hypothalamic-pituitary-adrenal axis, ACTH regulates responses to various stressors, including hypoglycemia, psychological stressors such as fear, and physical stressors such as hypovolemia. Upon binding to its receptor MC2R, ACTH acutely activates the intracellular cAMP-PKA pathway, thus increasing the expression of StAR12, which modulates the production of steroids. ACTH deficiency (ACTHD) is caused by disorders of corticotrophs or by disorders of the pituitary stalk or hypothalamus in the case of CRH deficiency and leads to nausea/vomiting, decreased appetite, hypoglycemia, and fatigue.
