Because baseline circulating IgE in naïve mice is exceptionally low compared to other immunoglobulins — often in the low ng/mL range — and rises dramatically during allergic sensitization or helminth infection, accurate quantification across this dynamic range demands both high sensitivity and appropriate assay calibration.
Detection Range: The 15.6–1000 pg/mL range captures IgE levels in both unsensitized and mildly sensitized mice without extensive sample manipulation, though serum from strongly challenged animals (e.g., chronic asthma or repeated OVA sensitization models) will likely require dilution, as total IgE can reach µg/mL concentrations.
Sensitivity: At 7.8 pg/mL, the kit detects the trace IgE present in naïve or antigen-specific IgE-depleted mice, enabling baseline measurements that many less sensitive assays miss entirely.
Sample Compatibility: Validation in both serum and plasma allows researchers to use the same assay across terminal bleeds and serial longitudinal sampling via submandibular or tail vein collection with anticoagulant-treated tubes, critical for tracking IgE kinetics within individual animals over sensitization protocols.
Assay Efficiency: The 50–100 µL sample volume and 1–5 hour completion time accommodate the limited blood volumes obtainable from mice, particularly in repeated-measures designs, while enabling same-day results across large cohorts.
