Epigen is the most recently identified ligand of the EGF receptor and plays a distinctive role in epithelial proliferation and tumor biology, making it a valuable tool for dissecting EGFR signaling specificity. This tag-free recombinant human EPGN, spanning residues 24–95 of Isoform 2, encompasses the EGF-like domain required for receptor engagement and demonstrates confirmed mitogenic activity with an ED50 below 300 ng/mL in Balb/c 3T3 proliferation assays, corresponding to a specific activity exceeding 3.3 × 10³ IU/mg. The E. coli expression system yields a non-glycosylated protein that isolates the intrinsic receptor-binding potency of the EGF-like domain from glycan-mediated effects, providing a suitable basis for receptor-ligand binding studies via SPR or ELISA, cell proliferation and survival assays, and in vivo tumor biology models. Purity exceeding 98% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based functional assays and antibody characterization workflows.
