The IL-17A/F heterodimer plays a distinct role in mucosal immunity and inflammatory signaling, bridging IL-17A and IL-17F homodimer activities through preferential engagement of the IL-17RA/RC receptor complex. This recombinant human IL-17A&IL-17F heterodimer (aa 24–155 of IL-17A & aa 31–163 of IL-17F, C-terminal 6×His-tagged) demonstrates picomolar affinity for IL-17RA (KD = 8.6 pM by BLI), confirming native-like receptor engagement suitable for NF-κB and MAPK signaling pathway studies, cell-based activation assays in synoviocytes or epithelial cells, and antibody development campaigns requiring a physiologically relevant coating antigen. The protein also binds IL-17RA-Fc with a KD of 10.3 nM, supporting its use as a reference standard in receptor-binding inhibition assays and neutralization screening. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for in vivo inflammation models and primary immune cell experiments where LPS-driven artifacts must be excluded.
