IL-36β (IL-1F8) signals through the IL-36 receptor to activate NF-κB and MAPK pathways, playing a central role in epithelial and immune cell inflammatory responses. This tag-free recombinant human IL-36β, spanning residues 5–157 of Isoform 2, faithfully represents the processed mature sequence and demonstrates strong signaling potency with an ED50 below 10 ng/mL in an IL-8 secretion assay using human preadipocytes, corresponding to a specific activity exceeding 1.0 × 10⁵ IU/mg. These activity data support use in cell-based functional assays measuring NF-κB or MAPK activation, downstream cytokine release studies, in vivo inflammation models, and as a quantitative reference standard in cytokine detection platforms such as ELISA. Endotoxin levels below 1.0 EU/μg minimize LPS-driven artifacts in sensitive immune cell experiments, while purity greater than 97% by SDS-PAGE provides a suitable basis for antibody development workflows including immunization and positive control applications.
