IL-4Rα is a critical signaling hub in type I and type II IL-4/IL-13 receptor complexes, making its extracellular domain (aa 26–231) an essential tool for dissecting Th2-driven immune pathways. This C-terminal Fc-tagged construct, expressed in mammalian cells to preserve native glycosylation and conformational integrity, demonstrates robust ligand engagement: immobilized IL-4Rα-Fc binds biotinylated human IL-4 with an ED50 of 2.43 ng/mL in functional ELISA, and it inhibits IL-4-dependent proliferation of TF-1 cells with an ED50 of 5–20 ng/mL, confirming biologically relevant blocking activity. These validated activities provide a suitable basis for receptor-ligand interaction assays by ELISA, SPR, or BLI, competitive inhibition studies, blocking antibody screening, and therapeutic antibody epitope mapping. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based neutralization assays and sensitive drug discovery screening campaigns.
