Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

IL-4Rα is a critical signaling hub in type I and type II IL-4/IL-13 receptor complexes, making its extracellular domain (aa 26–231) an essential tool for dissecting Th2-driven immune pathways. This C-terminal Fc-tagged construct, expressed in mammalian cells to preserve native glycosylation and conformational integrity, demonstrates robust ligand engagement: immobilized IL-4Rα-Fc binds biotinylated human IL-4 with an ED50 of 2.43 ng/mL in functional ELISA, and it inhibits IL-4-dependent proliferation of TF-1 cells with an ED50 of 5–20 ng/mL, confirming biologically relevant blocking activity. These validated activities provide a suitable basis for receptor-ligand interaction assays by ELISA, SPR, or BLI, competitive inhibition studies, blocking antibody screening, and therapeutic antibody epitope mapping. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based neutralization assays and sensitive drug discovery screening campaigns.

Receptor for both interleukin 4 and interleukin 13. Couples to the JAK1/2/3-STAT6 pathway. The IL4 response is involved in promoting Th2 differentiation. The IL4/IL13 responses are involved in regulating IgE production and, chemokine and mucus production at sites of allergic inflammation. In certain cell types, can signal through activation of insulin receptor substrates, IRS1/IRS2.; Soluble IL4R (sIL4R) inhibits IL4-mediated cell proliferation and IL5 up-regulation by T-cells.
