The production of the KRT14 monoclonal antibody starts with the immunization of mice using a synthesized peptide derived from human KRT14. After that, B cells are isolated from the spleen of the mice and fused with myeloma cells to generate hybridomas. The resulting hybridomas are screened to select out those that only produce antibodies that are specific to KRT14. Once specific hybridomas are identified, they are cultured in the mouse's abdominal cavity. The KRT14 monoclonal antibodies are purified from mouse ascites through affinity chromatography using a specific immunogen. The purified KRT13 monoclonal antibody is a mouse monoclonal antibody that specifically reacts with human KRT14 protein and can be utilized for ELISA and IHC applications. KRT14 is specifically expressed in the basal cells of stratified epithelia, such as the epidermis of the skin.
The main function of KRT14 is to provide mechanical strength to the epithelial cells and protect them from mechanical stress, while also playing a role in cell signaling, adhesion, and migration. Mutations in the KRT14 gene have been associated with several skin disorders, including epidermolysis bullosa simplex, a condition characterized by skin blistering and erosion.
