CLEC4C (BDCA-2/CD303) serves as a key receptor on plasmacytoid dendritic cells that modulates type I interferon signaling, making it a compelling target for immunological research and therapeutic antibody development. This recombinant construct covers the extracellular C-type lectin domain (aa 45–213), providing the ligand-binding region critical for receptor-ligand interaction studies, and its binding activity has been confirmed by functional ELISA with an EC50 of 31.43–43.52 μg/ml against a human CLEC4C antibody. The N-terminal 6xHis-SUMO tag supports efficient purification and enhances solubility from the E. coli expression system, while the validated antibody-binding capacity supports use in competitive inhibition assays, blocking antibody screening, epitope mapping, and as a positive control in ELISA or SPR-based affinity characterization. Purity exceeding 90% by SDS-PAGE satisfies the quality thresholds commonly required for ligand-binding assays and drug discovery screening of biologic inhibitors targeting dendritic cell function.
