Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

Complement C5 occupies a pivotal branch point in the complement cascade, where its cleavage generates both the potent anaphylatoxin C5a and the membrane attack complex initiator C5b, making it a critical target for studying immune activation and therapeutic intervention. This full-length mature protein (aa 19–1676) demonstrates robust antibody binding with an EC50 of 1.867–2.192 ng/mL in functional ELISA and a binding affinity of 0.285 nM by surface plasmon resonance, providing quantitative evidence that supports its use in complement activation assays, inhibitor screening campaigns targeting the terminal pathway, and as a positive control in complement-based ELISAs. The recombinant protein meets the purity and endotoxin criteria typical for hemolytic assays and opsonization studies, with >90% purity by SDS-PAGE and <1.0 EU/μg endotoxin, thresholds that minimize interference in sensitive functional readouts where even trace contaminants can trigger nonspecific complement consumption or cellular activation.

Function

Activation of C5 by a C5 convertase initiates the spontaneous assembly of the late complement components, C5-C9, into the membrane attack complex. C5b has a transient binding site for C6. The C5b-C6 complex is the foundation upon which the lytic complex is assembled.; Derived from proteolytic degradation of complement C5, C5a anaphylatoxin is a mediator of local inflammatory process. Binding to the receptor C5AR1 induces a variety of responses including intracellular calcium release, contraction of smooth muscle, increased vascular permeability, and histamine release from mast cells and basophilic leukocytes. C5a is also a potent chemokine which stimulates the locomotion of polymorphonuclear leukocytes and directs their migration toward sites of inflammation.
