LGR5 marks adult stem cells in multiple epithelial tissues and is overexpressed in colorectal and other cancers, making it a target of interest for both regenerative medicine and oncology antibody development. This full-length mature protein (aa 22–907) is expressed in mammalian cells to preserve the native glycosylation and disulfide bonding essential for proper GPCR folding, then presented on virus-like particles to mimic physiological membrane topology. Functional ELISA validation demonstrates specific antibody binding with an EC50 of 1.690–1.867 ng/mL, confirming that the extracellular leucine-rich repeat domain retains its native conformation and supports use in therapeutic antibody epitope mapping, competitive inhibition screening, and ligand-receptor interaction studies by ELISA or surface plasmon resonance.
