P-selectin mediates the initial tethering and rolling of leukocytes on activated endothelium and platelets, a critical step in inflammation and metastasis that has made it a therapeutic target in cancer and cardiovascular disease. This mammalian-expressed construct spanning residues 42–771 with the V640L variant retains native glycosylation and demonstrates quantified binding to a recombinant anti-SELP antibody with an EC50 of 3.120–3.578 ng/mL in functional ELISA and a KD of 0.956 nM by surface plasmon resonance, confirming that the extracellular domain adopts a conformation competent for ligand recognition. These binding parameters support use in selectin-ligand interaction studies, antibody development and epitope mapping, and cell adhesion assays investigating leukocyte rolling or tumor cell arrest on endothelial monolayers. Purity exceeding 95% by SDS-PAGE and endotoxin below 1.0 EU/μg meet the quality thresholds commonly required for functional cell-based assays and in vitro binding experiments.
