CD133 (Prominin-1) is a pentaspan transmembrane glycoprotein widely used as a surface marker for isolating cancer stem cells and hematopoietic progenitors, yet its five-transmembrane topology makes soluble recombinant production notoriously difficult. This construct presents the full-length mature protein (aa 20–865) in a virus-like particle (VLP) format expressed in mammalian cells, preserving native conformation and post-translational modifications critical for antibody recognition. Functional ELISA confirms robust binding activity: immobilized at 5 µg/mL, the protein binds an anti-PROM1 recombinant antibody with an EC50 of 0.43–0.72 ng/mL, supporting use as a positive control in antibody screening, specificity validation, and sandwich ELISA development. Endotoxin levels below 1.0 EU/µg, determined by LAL assay, satisfy criteria typical for cell-based binding studies and flow cytometry antibody titration experiments where lipopolysaccharide interference must be minimized.
