Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

R-spondin-1 amplifies canonical Wnt signaling by engaging LGR4/5/6 receptors and neutralizing Wnt inhibitors, making it a critical regulator of stem cell self-renewal and tissue homeostasis. This recombinant protein, spanning the complete mature form (aa 21–263) and expressed in mammalian cells to preserve native glycosylation, demonstrates robust potentiation of Wnt3a-driven Topflash reporter activity in CHO cells with an ED50 below 1.0 μg/ml, confirming functional receptor engagement at physiologically relevant concentrations. The sub-micromolar potency supports its use in stem cell expansion protocols, intestinal organoid culture systems, and mechanistic studies of Wnt pathway modulation where precise dose-response relationships are essential. Endotoxin levels at or below 10 EU/mg and purity exceeding 95% align with standards expected in cell-based differentiation assays and receptor-ligand binding experiments, including surface plasmon resonance and competitive ELISA formats designed to map LGR interaction kinetics.

Function

Activator of the canonical Wnt signaling pathway by acting as a ligand for LGR4-6 receptors. Upon binding to LGR4-6 (LGR4, LGR5 or LGR6), LGR4-6 associate with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. Also regulates the canonical Wnt/beta-catenin-dependent pathway and non-canonical Wnt signaling by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway. Acts as a ligand for frizzled FZD8 and LRP6. May negatively regulate the TGF-beta pathway. Has a essential roles in ovary determination. Regulates Wnt signaling by antagonizing DKK1/KREM1-mediated internalization of LRP6 through an interaction with KREM1.
