The recombinant Candida albicans ALS3 protein is a fusion protein consists of the Candida albicans ALS3 protein (918-1119aa) partnered with the N-terminal 10xHis-B2M tag and C-terminal Myc tag. It was produced in the E.coli. This recombinant ALS3 protein's purity is greater than 90% determined by SDS-PAGE. After electrophoresis, there is a 38 kDa protein band presented on the gel.
Candida albicans is part of the normal human flora, and it grows on mucosal surfaces in healthy individuals. Resistance caused by the formation of the Candida albicans (C. albicans) biofilm is one of the main reasons for antifungal therapy failure. Thus, it is important to find indicators that predict C. albicans biofilm formation to provide evidence for the early prevention and treatment of the C. albicans biofilms. ALS3 as a member of the agglutinin-like sequence (ALS) gene family, it was differentially expressed in free, antifungal, drug-sensitive C. albicans. A larger number of tandem repeats is associated with increased adherence when different-size alleles are expressed in C. albicans, but not when they are expressed in S. cerevisiae. ALS3 mediates adherence to diverse host substrates. ALS3 also binds to host cell ferritin and enables C. albicans to utilize this protein as a source of iron. Because of its multiple functions and its high expression level in vivo, Als3 is a promising target for vaccines that induce protective cell-mediated and antibody responses.
