Buffer

If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose.

Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

To generate biotinylated recombinant chicken glyceraldehyde-3-phosphate dehydrogenase (GAPDH) in E.coli, CUSABIO transfers the open reading frame (ORF) of the GAPDH of interest (2-333aa) into a plasmid that contains both the MBP-tag and 6xHis-Avi-tag biotinylation sequence, at the N- or C- terminus of the ORF, respectively. Next, the recombinant plasmid vector is introduced into the E.coli cells and expressed in E. coli with the pBirA Cm^r plasmid present. After that, the E.coli cells are harvested by centrifugation and lysed to release the cellular content, including the biotinylated recombinant chicken GAPDH protein, which is purified from the cell lysate, with up to 90% purity as accessed by SDS-PAGE.

Function

Has both glyceraldehyde-3-phosphate dehydrogenase and nitrosylase activities, thereby playing a role in glycolysis and nuclear functions, respectively. Glyceraldehyde-3-phosphate dehydrogenase is a key enzyme in glycolysis that catalyzes the first step of the pathway by converting D-glyceraldehyde 3-phosphate (G3P) into 3-phospho-D-glyceroyl phosphate. Participates in nuclear events including transcription, RNA transport, DNA replication and apoptosis. Nuclear functions are probably due to the nitrosylase activity that mediates cysteine S-nitrosylation of nuclear target proteins such as SIRT1, HDAC2 and PRKDC.

Results show that despite being commonly used housekeeping genes neither GAPDH mRNA nor beta-actin mRNA is expressed in all cell types or tissues at high levels. Strikingly, expression levels are very low in some organs. PMID: 23099774
